Endocrine control of renal function

Hormones regulate urinary excretion of water and ions by modulating the expression and activity of transmembrane tranporters (channels, carriers , pumps). Hormones belong to two families, those acting via membrane bound receptors and those acting via intracellular receptors.

Peptide hormones and cathecholamines generally act through receptors that are integral membrane proteins, have seven spanning domains, and are coupled through G proteins to intracellular second messengers. Binding of the hormones to the receptors releases intracellular second messengers that generally activate protein kinases and protein phosphatases. Reversible protein phosphorylation of target proteins is now generally recognized as a widespread mechanism by which external stimuli induce fast cellular responses. The adrenocorticoid and the thyroid hormones belong the family of hormones acting via intracellular receptors. When activated by the ligand, the receptor pass the nuclear membrane and bind to specific response elements on the promoter of target genes, interacting with the transcriptional machinery, and therefore modulating the expression of the target gene.

A proper effect of the hormones on renal function requires therefore the presence of a variety of different proteins (receptors, intracellular signalling messengers, effector proteins), whose expression may change during ontogeny. This lecture will particularly review the developmental aspect of the endocrine regulation of renal function.
